Laboratory for Orthopaedic Tissue Engineering and Regeneration

The University of California at San Francisco Laboratory for Orthopaedic Tissue Engineering and Regeneration is directed by Jeffrey C. Lotz, Ph.D. The laboratory is located within the Department of Orthopaedic Surgery (Parnassus campus) and is devoted to conducting basic research in several areas of orthopaedics. Recently, we have focused on understanding the etiology of different diseases (e.g., disc degeneration, osteonecrosis) and comorbidities (disc degeneration and diabetes). In the area of regenerative medicine, we are exploring various uses of mesenchymal stem cells for new therapies for disc, cartilage, and bone regeneration. Our diverse research team includes bioengineers, biologists, biochemists, histologists, and orthopaedic surgeons.


  • Cell Biology/Mechano-Biology: established cell lines, 3D cultures and different scaffolds, compression or tension-loaded in vitro bioreactor), RNA and DNA extraction and isolation, gene array and sequencing, ELISA assays, protein extraction, protein  detections (Western blot), PCR, and analysis of DNA, proteoglycan, and collagen content
  • In Vivo Modeling: rat tail disc degeneration model for device and drug testing; in-house small animal facilities and collaborations with large animal facilities at UC Davis, ISIS,& LyChron
  • Histology: Paraffin histology of soft tissue and decalcified bone, immunohistochemistry, glycomethlmethacrylate embedding, qualitative and quantitative histology analysis, and large-volume sectioning and staining
  • Surgical Samples: access to and storage of surgical samples; gene array analysis, proteomics, matrix quantification, and metabolomics
